In nature, approximately five hundred amino acids are found. Twenty of these are amino acids that serve as building blocks for proteins in the human body, referred to as 'fundamental amino acids'. These fundamental amino acids can be further divided into the categories 'essential', 'semi-essential', and 'non-essential'. L-arginine belongs to the so-called semi-essential amino acids, meaning that the body can partially produce it itself and to meet daily needs, it is partly dependent on diet. Adults can synthesize arginine from the amino acids ornithine, glutamine, glutamate, and proline; nonetheless, the amount of arginine in food significantly contributes to the total arginine supply. For children and adolescents, arginine is an essential amino acid because their biosynthesis is still too low.

L-arginine is incorporated into proteins and can be converted into other amino acids (such as ornithine and proline). Additionally, arginine is the only precursor of nitric oxide (NO), a biochemical signalling molecule. Arginine is also a precursor of creatine and polyamines.
